Si quelqu'un veut faire un film sur ma vie, vous n'avez qu'à me contacter. Contrairement au Dr. Magnanti, je n'ai rien à vous raconter mais on pourrait peut-être faire comme ces films français où il ne se passe rien pendant deux heures mais que les critiques aiment quand même.

Je ne devrais pas trop me casser la tête car, de toute façon, l'Hallowe'en n'est pas trop célébré là où j'habite. Pour ceux qui vont faire la fête ou passer de porte à porte, attention aux abus de bonbons!

wetbench_39 - Organismes synthétiques et transposons

(*) Des scientifiques du J. Craig Venter Institute développent de nouvelles techniques qui permettraient de créer un génome bactérien de toute pièce. Ces micro-organismes seraient ainsi complètement synthétiques.

Par ailleurs, une découverte parue cette semaine dans la revue Nature nous révèle que 74% du matériel génétique du mildiou de la patate, maladie à l'origine de la famine et de l'exode irlandaise au 19e siècle, est composé d'éléments transposables. En gros, près de 3/4 du génome de ce pathogène est composé de 'cochonneries' génétiques répétitives, énergétiquement très coûteux à maintenir. Pire, ces séquences sont instables et sont donc sources de mutations! Alors pourquoi les conserver? Les transposons peuvent certes être la cause d'effets délétères mais, chez le mildiou, il semblerait que les changements qu'ils génèrent contribuent à déjouer les défenses de l'hôte. Sans ces transposons, il n'y aurait peut-être pas de défilés annuels de la Saint-Patrick à New York ou sur la rue Sainte-Catherine à Montréal!

Mais pour revenir aux bibittes artificielles des chercheurs du JCVI, s'ils construisaient deux populations de bactéries complètement identiques à l'exception que l'une contiendrait des transposons et que l'autre en serait dépourvue, laquelle se porterait mieux, à terme et dans un environnement changeant?

In the news recently, scientists at the J. Craig Venter Institute find methods that should allow for the synthesis of living organisms, from scratch.

Another story published this week in Nature reveals that the genome of potato blight - the disease held responsible for the Irish famine and subsequent exodus in the 19th century - is mostly (3/4) made up of transposons. These are repeated genetic 'crap' that are energetically costly to maintain and that are highly unstable. Thus, they can be the cause of deleterious genetic mutations. So why keep these elements? Well, mutations are changes, and in the case of potato blight, change seems to be important for the pathogen to outsmart host defenses. Imagine: without transposons, there may not be St-Patrick's day parades in New York or on Montreal's Sainte-Catherine street!

So, getting back to the synthetic bugs of the JCVI researchers, if they were to synthesize two populations of organisms, one with and the other without transposons, which one would fare better in the long-run and in a changing environment?

Voici un outil publique du gouvernement américain pour visualiser les protéines en 3D.

Un peu de la même manière que le projet de recherche d'intelligence extra-terrestre SETI@home, Folding@home est un système distribué qui emprunte la puissance de votre ordinateur, lorsque vous ne l'utilisez pas, pour calculer des repliements proteiques. Ces opérations complexes demandent beaucoup de temps de calcul, alors vous pouvez mettre à contribution votre processeur en téléchargeant une petite application sur le site du projet Folding@home.

Here's a publicly available tool from the U.S. government that will allow you to visualize 3D structures of proteins.

Somewhat like the Search for Extra-Terrestrial Intelligence project SETI@home, Folding@home is a distributed computing system that borrows your computer, when you are not using it, to calculate protein folding. These complex operations require a lot of computing time and power so you can contribute your processor to the project by installing a little program that you can download from the Folding@home web site.

(1) Les nouvelles du mois de juin tournent autour du masque:
En politique française, le décret 'anti-cagoules' interdit désormais de se couvrir le visage lors de manifestations publiques;
Côté spectacles, décès de Michael Jackson, un adepte notoire du port du masque (je pense que je n'ai pas besoin d'indiquer un lien à la nouvelle);
Autour de la médecine, de la science et des politiques sanitaires, la grippe A(H1N1) passe en phase 6 du niveau de pandémie. Vous pouvez analyser vous-même les séquences du virus sur le site du NCBI.
Bref, il faut éviter de faire grève cet automne en France (foule sans masque en période de pandémie - le roi du pop n'a plus à se soucier de ces problèmes).
